ABSTRACT

@#Objective    To analyze the correlation between folate receptor-positive circulating tumor cells (FR+CTC) and the benign or malignant lesions of the lung, and to establish a malignant prediction model for pulmonary neoplasm based on clinical data, imaging and FR+CTC tests. Methods    A retrospective analysis was done on 1 277 patients admitted to the Affiliated Hospital of Qingdao University from September 2018 to December 2019, including 518 males and 759 females, with a median age of 57 (29-85) years. They underwent CTC examination of peripheral blood and had pathological results of pulmonary nodules and lung tumors. The patients were randomly divided into a trial group and a validation group. Univariate and multivariate analyses were performed on the data of the two groups. Then the nomogram prediction model was established and verified internally and externally. Receiver operating characteristic (ROC) curve was used to test the differentiation of the model and calibration curve was used to test the consistency of the model. Results    Totally 925 patients suffered non-small cell lung cancer and 113 patients had benign diseases in the trial group; 219 patients suffered non-small cell lung cancer and 20 patients had benign diseases in the verification group. The FR+CTC in the peripheral blood of non-small cell lung cancer patients was higher than that found in the lungs of the patients who were in favorite conditions (P<0.001). Multivariate analysis showed that age≥60 years, female, FR+CTC value>8.7 FU/3 mL, positive pleural indenlation sign, nodule diameter, positive burr sign, consolidation/tumor ratio<1 were independent risk factors for benign and malignant lung tumors with a lesion diameter of ≤4 cm. Thereby, the nomogram prediction model was established. The area under the ROC curve (AUC) of the trial group was 0.918, the sensitivity was 86.36%, and the specificity was 83.19%. The AUC value of the verification group was 0.903, the sensitivity of the model was 79.45%, and the specificity was 90.00%, indicating nomogram model discrimination was efficient. The calibration curve also showed that the nomogram model calibration worked well. Conclusion    FR+CTC in the peripheral blood of non-small cell lung cancer patients is higher than that found in the lungs of the patients who carry benign pulmonary diseases. The diagnostic model of clinical stage Ⅰ non-small cell lung cancer established in this study owns good accuracy and can provide a basis for clinical diagnosis.

ABSTRACT

Objective:To explore the effects of smoking on lung function and physical fitness of college students.Method:168 college students on September 30, 2020 were selected as the research subjects by convenient sampling. According to whether they had been smoking for more than 3 years in the past or (and) now, they were divided into smoking group (66 cases, 39.3%) and non-smoking group (102 cases, 60.7%). The general demographic data, body composition, health fitness and lung function of all the included subjects were collected, and the health status of the two groups were compared, the relationship between lung function and healthy physical fitness in the smoking group was analyzed, and the influencing factors of forced vital capacity (FVC), forced expiratory volume in the first second (FEV 1) and expiratory vital capacity (EVC) were also analyzed. Results:The peak expiratory flow rate (PEF) of male college students in smoking group was lower than that of non-smoking group [(9.37±1.72) vs (10.04±1.69) L/s] ( F=5.051, P=0.026). The standing time with eyes closed and one foot of smoking group was longer than that of non-smoking group [(87.2±49.1) vs (63.6±39.7) s] ( F=9.327, P=0.003). The number of sit ups per minute of female college students in smoking group was less than that in non-smoking group [(39.5±7.5) vs (49.2±12.5) times/min] ( F=5.189, P=0.029). The peak inspiratory flow rate (PIF), FVC, FEV 1, EVC and PEF in the smoking group were positively correlated with grip strength, back muscle strength, number of sit ups, push ups, standing high jump and standing long jump ( P<0.001); through the analysis of relevant data, the estimation formula of lung function and health fitness items was obtained. Conclusion:Smoking is harmful to college students′ lung function and physical fitness.

ABSTRACT

OBJECTIVE: To compare the quality of original preparations of Thioctic acid injection and generic preparations from 2 domestic manufacturers, discuss the acute toxicity test of mice and to investigate the project of consistency evaluation methods. METHODS: According to the quality standard that stated in Chinese Pharmacopeia, physicochemical properties (characters, pH, osmotic pressure, etc., contents and related substances of samples of Thioctic acid injection as well as LD50 in acute toxicity test (n=10), and mortality of mice after administration of injection solution (n=30) were compared among 3 manufacturers. RESULTS: The physicochemical properties as and related substances of the original drug and 2 generic drugs were all in line with the quality standard; the contents of 3 samples ranged 95%-105%. The acute toxicity test results showed that the LD50 values of 2 generic drugs (LD50: 247.911 mg/kg, 95% confidence interval: 222.209-277.999 mg/kg;LD50: 215.291 mg/kg, 95% confidence interval: 196.637-235.053 mg/kg) were smaller than that of original drug (LD50: 266.534 mg/kg, 95% confidence interval: 250.597-283.418 mg/kg), but there was no statistical difference (P>0.05). The results of 3 repeated experiments showed that there was statistical significance in the number of animal death caused by the 2 generic drugs (26, 28) was more than that of the original drug (19) (all P<0.05), when injection solution was injected into mice in a single dose. After administration of the original drug, mice showed excitatory reactions such as movement and squeal, while 2 generic drugs showed inhibitory reactions. CONCLUSIONS: 2 generic drugs of Thioctic acid injection and the original drug all conform to the relevant regulations of Chinese Pharmacopoeia in terms of preparation quality standards, but the results of acute toxicity test are quite different, so it is difficult to prove the consistency between the 2 generic drugs and the original drug. Therefore, acute toxicity test is necessary for the consistency evaluation of injections.

ABSTRACT

Traditional medicine (herb medicine) began to prevail again over last two decades, and it is about 70% of the world population taking herb medicine as supplement or alternative medicine according to a recent survey. The consumption of herb medicine increased exponentially in Canada, Australia and Europe during last 10 years. Since concomitant administration of herbal and western medicine has become a trend, it requires paying close attention to the problem. Herb-drug interactions have been extensively investigated worldwide, and there is an increasing concern about the clinical herb-drug interaction. In this review we introduced the current progress in the herb-drug interactions including evidence-based clinical studies and establishment of levels of evidence for herb-drug interaction; and in the related mechanisms including the induction and inhibition of metabolic enzymes, inhibition and induction of transport and efflux proteins, alteration of gastrointestinal functions, and alteration in renal elimination. We also analyzed both the achievements and the challenges faced in the concomitant administration of traditional Chinese medicine and western medicine.

ABSTRACT

The study is to establish an HPLC method using fluorescence detector for the determination of doxazosin enantiomers and investigate their chiral inversion in vitro and in vivo. Ultron ES-OVM was taken as the chiral chromatographic column, and the column temperature was 30 degrees C. Isocratic elution using a mobile phase of phosphate buffer-acetonitrile (85 : 15, v/v) at a flow rate of 0.8 mL x min(-1) was done. The fluorescence detection was set at lambda(Ex) = 255 nm and lambda(Em) = 385 nm. Prazosin was used as the internal standard. (-) Doxazosin or (+) doxazosin added into rat plasma in vitro was determined after incubating in 37 degrees C water bath for 2, 5 and 10 days. (-) Doxazosin or (+) doxazosin was administered orally to the rats for one months. Plasma samples were taken at 8 h after the last administration. A good linear relationship was achieved when the concentration of doxazosin enantiomers was within the range of 4 - 2 000 ng x mL(-1). The average recovery for (-) doxazosin was 99.5% with RSD 3.6%, and for (+) doxazosin was 99.3% with RSD 4.3%. Chiral inversion was observed neither in vitro nor in vivo studies. The method is selective, accurate and reproducible, which is suitable for the detection of doxazosin enantiomers in rat plasma. The in vitro and in vivo studies indicate that chiral inversion occurs uneasily between (-) doxazosin and (+) doxazosin in the rat.

ABSTRACT

A micellar electrokinetic capillary chromatography method with ultraviolet detection was developed for the simultaneous determination of psoralen, xanthotoxin, isoimpinellin, bergapten and scopoletin in Radix Glehniae. The separation was performed on an uncoated fused silica capillary column (50.2 cm x 75 microm x 40 cm) with 20 mmol x L(-1) borax solution (pH 9.6) containing 16 mmol x L(-1) sodium dodecylsulfate (SDS) and 15% acetonitrile as running buffer at applied voltage of 22 kV. The detection wavelength was 214 nm. The effects of concentrations of borax solution, sodium dodecylsulfate (SDS), and organic modifier, voltage, temperature on the separation and sensitivity were investigated. The five active constituents were completely separated within 7 min. The linear ranges of psoralen, xanthotoxin, isoimpinellin, bergapten and scopoletin were 9.91-82.6, 37.2-162, 2.23-18.6, 2.73-22.3 and 2.89-20.1 mg x L(-1), respectively. And the average recoveries were 98.9%, 98.4%, 101.3%, 99.1% and 98.0%, respectively. This simple and rapid method provided a new basis for assessment on quality of Radix Glehniae.

ABSTRACT

<p><b>OBJECTIVE</b>To develop a HPCE analysis method for fingerprints of Forsythia suspensa from Hebei province, get reference fingerprint and compare the fingerprints of F. suspensa collected from different producing areas and different parts of the plant.</p><p><b>METHOD</b>Electrophoresis was performed on a fused silica capillary column (75 microm x 60 cm, 30 cm). The running buffer was composed of 50 mmol x L(-1) borax (adjust to pH 9.90 with 0.1 mol x L(-1) NaOH). The applied voltage was 15 kV and the temperature was 20 degrees C. The detection wavelength was 214 nm. The semblances to the crude drugs of different producing areas were compared.</p><p><b>RESULT</b>The mutual mode of HPCE fingerprints was set up with 12 common peaks. The fingerprints of F. suspensa from Hebei province had high similarity, F. suspensa from Shanxi and Henan were also of good quality. The chemical composition in different parts of the herb had big differences.</p><p><b>CONCLUSION</b>The method is simple, quick, accurate and can be used as a new means for the quality control of F. suspensa.</p>

ABSTRACT

Objective To develop a sensitive, simple, and accurate method for the determination of shionone in rat plasma after ig administration of Asteris Radix petroleum ether extract (RAPE). Methods The separation was achieved by HPLC on a RP18 column (150 mm × 3.9 mm, 5μm) with a mobile phase composed of acetotitrile-0.05% phosphoric acid water (98: 2) at a flow rate of 1.0 mL/min. UV Detector was set at 200 nm and friedelin was chosen as an internal standard. Results The linear range of the standard curves was (0.3443-22.0) μg/mL with the correlation coefficient of 0.9968. The intra- and inter-day precisions were all below 10% and the relative error was -3.5%-1.1%.Conclusion The developed method can be successfully applied to the pharmacokinetic study. After ig administration of RAPE, T1/2(ka) is (33.09 ± 7.32) min and T1/2(ke) is (84.95 ± 22.34) min.
